I go on a walking path and I recently realized I could either scoop some of this little river water up or I found a little hole with some good looking ecosystem to it, I’ve been wanting to do this for a long time but I’m finally at a place where I actually could have one, so I have a couple questions, when I scoop the water up do I want to go deep enough to also get some dirt as well? Also do I want to add in a couple sticks from around the area as well that would fit in the jar? Are these good sources of water also?

Grab mud from surface/bank area. Then if you can get some from deeper. Same with the water.
Glad to see more jarrarium hobbyists do this sort of thing. Do give us an update once you set it all up!
