Definitely not. That’s where the plant will get energy to produce more leaves. If you don’t like the placement of it, wait until it has 3-4 more big leaves before removing it.
I’ve seen propagation advice that you can cut the big leaf in half so it can still provide energy but not need to use as much energy to sustain the leaf. But I’ve not tried it myself so might be worth looking into
