I’ve wanted a greenhouse for years and I keep coming across those DIY angled brackets that use 2×2 lumber. Has anyone successfully put one of these together? What did you use for paneling when finished? How strong are they?

It would probably help if you linked what you are talking about.
If you went to a framing factory with a plan, they could run up a frame for you with metal gang nails. You would just need to add purlins. Those plastic brackets would have a limited lifespan, eventually they would get brittle.
